Enter the measured OD660 and the dilution factor used in the photometer. The algorithm applies a polynomial correction to account for the non-linear relationship between optical density and actual cell numbers caused by light scattering, ensuring a standardized stock of 3.2 × 106 cells/ml.
If you used 200 μl culture in a 1000 μl total volume (1:5 dilution), enter a dilution factor of 5.
Use this suspension as the Master Stock for 1:5 serial dilutions. Spotting 3.5 μl results in mathematically three cells in the final (6th) dilution.
